Is it possible to define an area map in a mod_slideshow slide? We have an image with different “hot” areas, how can we define them and display the slide within the module?

Sorry but that is not possible with JA Slideshow 2. It simply calls and animates the images based on certain variables. There may be other components available on the JED that fit your needs better. I am not aware of any though.
