I had the same problem in the earlier version of JA healthcare. Then i read somewhere that there was a bug and it was fixed in the latest version. The bug is still there..

1. In the top menu click Expo. – IT WORKS.
2. Under expo click ‘universities online exhibition’ – IT WORKS
3. Now click ‘Expo’ and IT DOESN’T WORK. (yet the ‘expo’ link in the breadcrumbs DOES WORK).

I had the same problem before. Doesn’t matter what menu items the top parent menu item has under it, when they are clicked, the top parent always gets disabled.

I created test menu under Expo menu and it’s working well when you click back on Expo menu and the problem on your site comes only from Universities Online Exhibition, Colleges Online Exhibition, Schools Online Exhibition, Training, Short Courses, CPD and Apprenticeship Expo menus (it’s SobiPro component).

In this case, you need to contact SobiPro component and ask them all menu items with Menu Item Alias menu type by Joomla default is disabled. Hope you will get best answer from them.

The second of your question, I see the configuration from backend of your site is working with LESS files as my highlight, you need to change Development Mode to OFF and it will work with the CSS files and you can re-define the new background with new theme under Theme tab
